When comparing enzyme supplements, one of the most important distinctions is the difference between how much enzyme material is present and how much measurable enzyme activity it provides.

These are not the same thing.

A product label may identify an enzyme in milligrams (mg), activity units, or both. Understanding the difference can make enzyme labels considerably easier to interpret.

Milligrams Measure Weight

A milligram is a unit of mass.

If a label states that a serving contains 100 mg of an enzyme preparation, the 100 mg describes the weight of that material.

It does not, by itself, tell you how much enzymatic activity the preparation provides.

Conversely, two products could provide the same number of FU using different amounts of ingredient if the starting materials have different activity concentrations.

Therefore:

mg = amount by weight

while

FU = measured fibrinolytic enzyme activity under the applicable assay

These measurements describe different characteristics of the ingredient.

FU Per Gram

Bulk nattokinase ingredients are frequently specified according to their activity per gram.

For example, an ingredient specification might state:

20,000 FU per gram

or

100,000 FU per gram.

If an ingredient is standardized at 20,000 FU per gram, then mathematically:

1 gram = 20,000 FU

500 mg = 10,000 FU

100 mg = 2,000 FU

This calculation assumes that the ingredient actually meets its stated activity specification.

That is where analytical testing and the Certificate of Analysis become important.

Why the Certificate of Analysis Matters

A Certificate of Analysis, commonly called a COA, provides analytical information about a specific ingredient or production lot.

For an enzyme ingredient, relevant information may include:

  • lot or batch number
  • enzyme identity
  • activity specification
  • activity test result
  • analytical method
  • microbiological testing
  • other applicable purity or quality specifications

The exact information provided depends on the ingredient, manufacturer and testing program.

When evaluating an enzyme ingredient, the stated weight and stated activity should therefore be considered separately.

Activity Is Not the Same as Clinical Effect

Another important distinction is that enzyme activity measurements are analytical measurements, not automatic measurements of clinical efficacy.

For example, a higher FU value indicates greater measured fibrinolytic activity according to the applicable assay.

It does not, by itself, establish that a product prevents, treats or cures a disease.

Clinical outcomes require appropriate clinical evidence.

How to Compare Enzyme Supplements

When comparing enzyme products, consider more than the number of milligrams printed on the front of the label.

Look for:

Enzyme identity
What enzyme is actually being supplied?

Ingredient amount
How much material is present?

Activity specification
Is enzymatic activity stated?

Activity unit
What measurement system is being used?

Activity per serving
How much measured activity does the serving provide?

Testing
Is the activity specification supported by appropriate analytical testing?

Lot documentation
Is batch-specific documentation available when appropriate?

These questions provide substantially more information than ingredient weight alone.

Nattokinase is an especially useful example because consumers frequently encounter products labeled in both milligrams and FU.

A milligram value tells you the weight of the nattokinase preparation.

An FU value provides information about its measured fibrinolytic activity under the applicable assay.

Milligrams and enzyme activity units measure different things.

Milligrams measure weight.
